ATF-SUBmerged™ kit with 430 cm2 HFF to fit the 4 liter VV B-SUB
The 4 liter Vessel Volume B-SUB (Batch-Single-Use-Bioreactor) is equipped with a ID19 mm / PG21 head-plate port closed with a plug and thus able to operate as both a Batch-SUB or Perfusion-SUB. When this unique B-SUB needs to operate as a P-SUBmerged the "CellRetention-PG21-kit-x" is easily inserted though the ID19 mm port under the hood by end-user. Remember to pre-wett the HFF and ready to use.
FlowChart connects the P-SUBmerged to the more advanced Cronus-PCS running Lucullus SCADA software. Rendered CAD illustrating the CellRetention-4 B-SUB enclosed in SUJ with liquid connections, all the hoses needed for PCS connections.
Inside the B-SUB:
- Aeration by super-efficient 25 or 100 µm pore glass micro sparger for high cell density, deep-tubes, addition, harvest, baffles, etc – see doc at download
- Optional CellRetention-PG21-kit-x DYI insert - SepraPor XFCM20C112-xx1x HFF (Hollow-Fibre-Filter) with 420 cm2 filter surface area and HFF inlet SUBmerged directly into the broth with inverted Thalia TM80 SUP on top
- Agitation with triple blade Kidney OD 60 mm, 45° blade angled
- Single-Use-Sensors (SUS) for DO, pH, bio-mass, temp, level as needed
Outside the B-SUB:
- HPD (Head-Plate-Drive) or MBD (Magnetic-Bottom-Drive) - check under Commerce
- 900 mm weldable M-Flex hose connected to various in & Out
- Thermal control by liquids - equipped with Single-Use-Jacket (SUJ) (InCyte not possible)
The B-SUB it consists of:
- 0.4 – 3.2 liter Working Volume (WV) from 4.0 liter Vessel Volume (VV) (dim OD150 x 270 mm) fully pre-assembled and ready to install with known impeller Power Numbers
- The CellRetention-4 is dual foil bag packed and precision E-beam irradiated

Benefits:

Installation (inside the hood):
- Release and remove the 19 mm plug from the PG21 port
- Inset the CellRetention-33-80C112-x kit elevated 20-25 mm above the bottom and tighten the nut with 1-2 Nm torque so the HFF is fixed tight
- Don’t forget to pre-wet HFF by adding suitable liquid and amount into the STR / SUB. Activate Clotho for 15 strokes and pump entire volume out through the permeate port by external peri pump

Two different "CellRetention-PG21-kit-x" delivered bagged and E-beam irradiated ready to be inserted in the 4 liter B-SUB through the ID19 mm port under the hood and thus become CellRetention P-SUBmerged. Both the Straight and the Angled Thalia is a Direct-Fit with the SepraPor C112-xx1x - 0.2 µm pore HFF with 420 cm2 surface. Choose Straight or Angled Thalia as needed from Commerce.
Clotho Drive Unit is required and integrates pressures, vacuum, volume sensing eliminating the need for external sensors for HFF DeltaP / ∆P pressure drop, mass-flow measurements. Clotho process software controls accurately the programmable stroke volume, number of strokes, the allowed pressure drop along the HFF for fully automatic broth low harvest velocity and fully selectable broth high velocity for the sequential lumen cleaning eliminating HFF blockage.
