0.4 – 2 liter ATF Perfusion-SUB

SUBmerged™ P-SUB keep the HFF free from fouling

This unique 3 liter bioreactor can operate as a batch SUB or as Perfusion-SUBmerged (P-SUB) after the ATF method

Benefits as P-SUB are:

  • HFF fouling – freedom to select broth velocities flushing out debris from inside the lumen
  • Dead volume – skip all the hoses and plastic connectors
  • Cell death - inverted Thalia pump above the HFF and P-SUB cannot accumulate cells
  • Shear force – remove all hoses and fittings and SUBmerge the HFF into the broth inside the P-SUB

SUBmerged 3 liter features:

  • 0.4 - 2 liter Working Volume fully pre-assembled P-SUB
  • Alternating broth by Thalia diaphragm Alternating-Single-Use-Pump (A-SUP) straight or angled version
  • SepraPor C112-xx1x HFF (Hollow-Fibre-Filter) with 420 cm2 surface SUBmerged directly into the broth

Left photo - The re-invented 3 liter P-SUB with SUBmerged SepraPor C112 and inverted Thalia TM80 with red LASER support on top. Centre photo - The InCyte biomass SUS patch seen mounted on the vessel wall and InCyte ARC sensor clicked on. Right photo - MST explain the 3 liter P-SUB is mounted in the Heating-Support-Foot (HSF) on the Magnetic-Stirrer-Table (MST). Reason is simple – there is not space for both servo motor, sensors and Thalia A-SUP on the OD137 mm head plate.

Industry leading architecture: P-SUBmerged integrates CellVessel SUB / A-SUP / HFF / SUS with:

  • 3.2 liter Vessel-Volume P-SUB offer 0.4 – 2 liter Working Volume (WV) in an OD 137 mm vessel
  • SUBmerged integrate the Thalia diaphragm Alternating-Single-Use-Pump (A-SUP) with the SepraPor HFF
  • SepraPor C112-xx2x with 0.2 µm pore, 55 x 1.0 mm PE straw/lumen, 420 cm2 area, Direct-Fit connection, one permeate side port
  • Ground-breaking Thalia LASER sensor guided A-SUP
  • Single-Use-Sensors (SUS) for DO, pH, bio-mass, temp, level or as needed

Rendered SolidWorks CAD drawings show different details of the SUBmerged 3 liter P-SUB all driven from bottom. Left P-SUB arranged in the Heating-Support-Foot on MST. Center the 3 liter P-SUB surrounded by the Re-Useable-Jacket (RUJ). Right the P-SUB incl SUJ on MST. Easy to spot the SepraPor SUBmerged HFF and the inverted straight angled Thalia SUP on top of the HFF.

Thermal control as needed – electrical or liquid with Magnetic-Bottom-Drive or Head-Plate-Drive. Electrical-Heating-Blankets or Heating-Support-Foot as options or the P-SUBmerged inserted into the Re-Useable-Jacket (RUS).

If Head-Plate-Drive (HPD) is the desired option – not the safest option for a lightweight P-SUB with a heavy servo motor on top – though if the P-SUBmerged require liquid thermal control, then by P-SUB inserted into the Re-Useable-Jacket (RUS) safety in obtained. The P-SUB version HPD, SUJ need to be equipped with the angled Thalia A-SUP allowing space for the servo motor.

Two different "CellRetention-3-kit-x" delivered bagged and E-beam irradiated ready to be inserted in the 3 liter CellVessel SUB through the ID19 mm port in the hood and thus become P-SUBmerged. The straight Direct-Fit Thalia ID19 mm tube part and the angled Direct-Fit Thalia connected to the SepraPor C112 HFF - as selected.

FlowChart connects the P-SUBmerged to the more advanced Cronus-PCS running Lucullus. Rendered CAD illustrate the 3 liter SUBmerged with straight Thalia and with angled Thalia.
