0.4 – 2 liter ATF Perfusion-SUB

SUBmerged™ P-SUB keep the HFF free from fouling

This unique 3 liter bioreactor can operate as a batch SUB or as Perfusion-SUBmerged (P-SUB) after the CellRetention ATF method.

Benefits of the P-SUBmerged are:

  • avoid HFF fouling – freedom to select broth velocities for gentle harvest and sequential pulses flushing out debris from inside the HFF
  • eliminate dead volume – skip all the hoses, traditional plastic connectors and SUBmerge the HFF
  • avoid cell death - inverted Thalia pump arranged on top of the HFF = hereby the diaphragm pump cannot accumulate cells
  • reduce shear force – protect cells by eliminating hoses, fittings - SUBmerge the HFF inside the P-SUB and the inverted Thalia on top of HFF

Centre photo - The InCyte biomass SUS patch mounted on the vessel wall and InCyte ARC sensor clicked on. Right photo - 3 liter P-SUB is mounted in the Heating-Support-Foot (HSF) on the Magnetic-Stirrer-Table (MST). Reason is simple – there is not space for both servo motor, sensors and Thalia TM80 on the OD137 mm head plate so Magnetic-Bottom-Drive (MBD) required.

P-SUBmerged 3 liter features:

  • 0.4 - 2 liter Working Volume (WV) fully pre-assembled and ready for use with selectable impellers of which Power Numbers are known
  • Thalia diaphragm Alternating-Single-Use-Pump (A-SUP) straight or angled version integrating a
  • SepraPor C112-xx1x HFF (Hollow-Fibre-Filter) with 420 cm2 surface SUBmerged directly into the broth
  • Single-Use-Sensors (SUS) for DO, pH, bio-mass, temp, level as needed

Rendered SolidWorks CAD drawings show different details of the 3 liter P-SUBmerged all 3 driven from bottom. Left P-SUB arranged in the Heating-Support-Foot on MST. Center the 3 liter P-SUB surrounded by the Single-Use-Jacket (SUJ) for liquid thermal control. Right the P-SUB incl SUJ on MST. Easy to spot the SepraPor SUBmerged HFF and the inverted straight angled Thalia SUP on top of the HFF.

Thermal control as needed - electrical or liquid with Magnetic-Bottom-Drive (MBD) or Head-Plate-Drive (HPD).

  • Electrical-Heating-Blankets (EHB) or electrical Heating-Support-Foot (HSF) as options
  • Liquid - the P-SUBmerged can be inserted into the Re-Useable-Jacket (RUS) or equipped with Single-Use-Jacket (SUJ)

If Head-Plate-Drive (HPD) is the desired option – not the safest option for a lightweight P-SUB with a heavy servo motor on top – though if the P-SUBmerged require liquid thermal control. Then by P-SUB inserted into the Re-Useable-Jacket (RUS) safety in obtained. The P-SUB version HPD, SUJ or RUJ need to be equipped with the angled Thalia A-SUP allowing space for the servo motor.

A unique option - batch or perfusion selectable from the same SUB

The 3 liter customizable CellVessel™ is available as B-SUB equipped with a ID19 mm port in the head plate and thus able to operate optionally as a Batch-SUB. When this unique B-SUB needs to operate as a P-SUBmerged the "CellRetention-3-kit-x" (pn 31-0021) is easily inserted though the ID19 mm port under the hood by end-user. And vupti a SUBmerged ATF 3 liter is created in minutes.

Two different "CellRetention-3-kit-x" available bagged and E-beam irradiated ready to be inserted in the 3 liter CellVessel B-SUB through the ID19 mm port in the hood and thus become CellRetention P-SUBmerged. Both the straight and the angled Thalia is a Direct-Fit with the SepraPor C112 HFF. Choose straight with MBD or or angled Thalia with HPD as needed from Commerce.

FlowChart connects the P-SUBmerged to the more advanced Cronus-PCS running Lucullus SCADA software. Rendered CAD illustrate the 3 liter SUBmerged with straight Thalia and with angled Thalia.
