Traditional 150 – 300 ml Working Volume P-SUB with A-SUP and HFF
Traditional ATF-External perfusion-SUB setup for Process-Development copying RefineTech concept from year 2000 with diaphragm pump below the standard HFF connected via Luer-Loc. Here combined with the Thalia-TM80 Single-Use-Pump (A-SUP) for alternating exchange of the broth flow along the 0.2 µm pore size Hollow-Fiber-Filter module (HFF).

Examples of standard P-SUBs ranging 250 – 300 ml Vessel Volume. All standard SUBs with no SUB modifications. Standard 80 cm2 HFF with Luer-Loc connections. This traditional setup face high velocities during harvest and potential high shear force stress as to the use of Luer-Loc. The SUP arranged below the HFF will accumulate cells and introduce a broth temperature drop.
Each shown P-SUB in cultivation control of its respective Process-Control-System (PCS). Modular 250 ml connects to its standard DO and pH sensor. The 300 ml BioBlu connects to DASbox pH and DO. The 500 ml CellVessel integrated Hamilton VisiFerm and OneFerm Single-Use-Sensors plus level sensors.
Example P-SUB cultivation controlled by Modular PCS. Drive of the Clio A-SUP is the Clotho drive unit. 500 ml CellVessel setup with Maxon servo motor for Head-plate-Drive with deep tube in one PG13 port and Luer-loc connectors. In this case show with head-plate drive and the P-SUB placed inside the Heating-Support-Foot. May also be driven via Magnetic-Bottom-Drive. May connect to a range of different PCS.
- The SUP exchange broth from the SUB through the HFF and return to the SUB through the HFF. Simultaneously the permeate pass the HFF barrier as harvest
- Thalia TM80 performance is guided by the re-usable Laser sensor
- The Laser sensor measures in real-time with high precision the position of the diaphragm for volume exchange and broth velocity calculation performed by Clotho
- Thalia TM80 can exchange any volume within its spec (Cardiac Output) and velocity from 0 to +10 m/s through the HFF
P-SUB – perfusion controlled by the Clotho Drive Unit:
- Drive Unit is designed to handle two Thalia SUPs and ensure efficient and reproducible operation
- Clotho easy programmable broth/retentate velocity and volume through HFF module for harvest and for removal of membrane deposits
- Connection to compact pressurized air and vacuum unit's like the Alagonia and Sarpedon - if the lab do not offer pressurized air and vacuum
